In vitro Expression Cloning Using the TNT® Coupled Reticulocyte Lysate System

The in vitro expression cloning or IVEC technique is a systematic and broadly applicable method for the genetic characterization and biochemical screening of genes and cognate proteins. IVEC uses the TNT® Coupled Reticulocyte Lysate System to express proteins from small pools of cDNA libraries, greatly simplifying the process of isolating genes that encode proteins identified by a particular biochemical function or characteristic. Here, we summarize the experimental procedure and suggest possible uses for the technology.
